The Effect of Two Years of Secukinumab Treatment on Bone Metabolism in Patients with Radiographic Axial Spondyloarthritis: Results from Daily Clinical Practice

Biologics 2023;17:161–166 doi: 10.2147/BTT.S434318

Siderius, et al. found that secukinumab was associated with low spinal radiographic progression. Furthermore, bone-related outcomes and BTMs related to collagen resorption (sCTX, PINP) remained constant during the 2-year period, whereas the BTM related to mineralisation (BALP) decreased significantly.

The Risk Factors for Uveitis Among Psoriatic Arthritis Patients: A Population-based Cohort Study

Clin Rheumatol. 2023;43(3):1053–1061 doi: 10.1007/s10067-023-06834-y

Retrospective matched cohort study concludes that PsA is associated with increased risk of uveitis, with past uveitis and treatment with etanercept being independently associated with a higher risk of uveitis. Patients who developed uveitis were mainly treated with topical glucocorticoids.

Risk of Major Adverse Cardiovascular Events in Immune-Mediated Inflammatory Disorders on Biologics and Small Molecules: Network Meta-Analysis

Clin Gastroenterol Hepatol. 2023 Oct 9:S1542-3565(23)00767-X doi 10.1016/j.cgh.2023.09.033 Epub ahead of print

The results of this study show that anti-IL-12/23, JAK inhibitors, and anti-TNF-α were associated with slightly higher risk of MACE compared with placebo. The risk was no different between biologic treatments, and the magnitude of risk did not differ between IMID type.

Upadacitinib in Patients with Psoriatic Arthritis and Inadequate Response to Biologics: 3-year Results from the Open-Label Extension of the Randomised Controlled Phase 3 SELECT-PsA 2 Study

Clin Exp Rheumatol. 2023;41(11):2286–2297 doi: 10.55563/clinexprheumatol/8l7bbk.

Data from this open-label extension showed the efficacy of upadacitinib observed at 56 weeks was maintained through to 152 weeks in the treatment of patients with PsA. No cumulative adverse effects were observed, and no new safety signals were identified.

Gender-specific Differences in Patients with Psoriatic Arthritis Receiving Ustekinumab or Tumour Necrosis Factor Inhibitor: Real-world Data

Rheumatology (Oxford). 2023 doi: 10.1093/rheumatology/kead089

These real-world data from PsABio on gender differences suggest that, at the start of biologic treatment, females have a worse clinical picture of PsA than males. In coming to these conclusions and others, this study sought to investigate effects of gender on disease characteristics and treatment impact in patients with PsA.

Clinical Efficacy of Alternative TNF Inhibitor and Secukinumab Between Primary Non-responder and Secondary Non-responder of Prior TNF Inhibitor in Ankylosing Spondylitis

Mod Rheumatol. 2023 doi: 10.1093/mr/roac005

This study demonstrated comparable drug retention between AS patients treated with alternative TNFi and secukinumab after failing to respond to prior TNFi therapy. The objective of this study was to compare the drug retention times and clinical efficacy of alternative TNFi and secukinumab in primary and secondary
non-responders with AS.

Risk of Hospitalization for Serious Infection After Initiation of Ustekinumab or Other Biologics in Patients with Psoriasis or Psoriatic Arthritis

Arthritis Care Res (Hoboken) 2022 doi: 10.1002/acr.24630

In this investigation it was concluded that other biologics and apremilast were associated with a 1.4- to 3-times higher risk of hospitalisation for serious infections in PsO/PsA patients when compared to ustekinumab. These findings should be considered in the safety profile of these therapies when selecting appropriate treatment regimens in patients with PsO/PsA.

Certolizumab Pegol Efficacy in Patients with Non-Radiographic Axial Spondyloarthritis Stratified by Baseline MRI and C-Reactive Protein Status: An Analysis From the C-axSp And Study

ACR Open Rheumatol. 2022 doi: 10.1002/acr2.11469

The results of this analysis indicated that certolizumab pegol (CZP) treatment benefits patients with nr-axSpA across all the MRI/CRP subgroups studied. To reach this conclusion, this study evaluated clinical responses to CZP in patients with nr-axSpA stratified by baseline MRI/CRP status.

Persistence and Effectiveness of the IL-12/23 Pathway Inhibitor Ustekinumab or Tumour Necrosis Factor Inhibitor Treatment in Patients with Psoriatic Arthritis: 1-year Results From the Real-world PsABio Study

Ann Rheum Dis 2022 doi: 10.1136/annrheumdis-2021-221640

Many RCTs have demonstrated efficacy and safety of biologics in PsA. However, long term comparative real world data is lacking. This study aimed to evaluate the real-world effectiveness and persistence of the IL-12/23 inhibitor ustekinumab or a TNFi for PsA 1 year post initiation. As a result, they found that PS-adjusted comparisons demonstrated comparable overall persistence, effectiveness and safety for both modes of action in PsA.

Comparison of drug retention of TNF inhibitors, other biologics and JAK inhibitors in RA patients who discontinued JAK inhibitor therapy

Rheumatology (Oxford) 2022 doi: 10.1093/rheumatology/keac285

Real-world population-based study shows that a switch to a second JAKinib results in a higher drug retention, as compared to switching to a TNFi, in patients with RA who discontinue original JAKinib therapy.
