Gollins et al. reported that within this cohort, the Psoriatic arthritis response criteria (PsARC) response to 4th+ lines of b/tsDMARD was not significantly reduced compared with 2nd/3rd line in participants who had failed at least 3 b/tsDMARDs. Authors evaluated the primary clinical response to sequential lines of b/tsDMARD therapy in PsA, focusing on the effectiveness of later line treatments.

Mease et al. report that patients without radiographic progression through 2 years of secukinumab treatment had greater achievement of LDA states at Week 104 than patients with radiographic progression. This post hoc analysis by Mease et al. of the FUTURE 5 study evaluated the relationship between radiographic progression status at Week 104 and achievement of LDA or remission and identified demographics and clinical characteristics that were associated with radiographic progression status at Week 104.

Mease et al. showed that guselkumab provided significantly higher rates of clinical improvement and significant inhibition of structural damage progression versus PBO, with no new safety signals, at Week 24 in biologic-naïve participants with active and erosive PsA. Mease et al. report primary efficacy and safety results for the double-blind PBO-controlled phase (Weeks 0-24) of the 3-year APEX study.

In this first global clinical study of a nanobody in inflammatory arthritis, sonelokimab, an
IL-17A- and IL-17F-inhibiting nanobody demonstrated strong efficacy across multiple domains including high-hurdle composite joint and skin responses. McInnes et al. reported on the Phase 2, randomized, double-blind, PBO-controlled ARGO trial which evaluated the efficacy and safety of sonelokimab in patients with active PsA.

Treatment with risankizumab provides durable improvement in the signs and symptoms of PsA across all the GRAPPA disease domains and related conditions. This post hoc analysis by Coates et al. aimed to assess the efficacy of long-term treatment with risankizumab across the updated GRAPPA domains and key related conditions of PsA.

McInnes et al. assessed the efficacy of guselkumab over 48 weeks in patients with psoriatic arthritis who had an inadequate response to TNF inhibitors. The results demonstrated consistent improvements in joint, skin, and patient-reported outcomes across all baseline-defined subgroups. Guselkumab showed greater efficacy compared with placebo at Week 24, with responses maintained or improved through Week 48.

Kavanaugh et al. conducted a post hoc analysis of a phase 2 trial examining deucravacitinib in patients with active psoriatic arthritis, focusing on the achievement of MDA components.

The analysis demonstrated that treatment with deucravacitinib led to a higher proportion of patients meeting each MDA component compared with placebo at 16 weeks.

Compared with placebo, bimekizumab-treated patients displayed a rapid clinically meaningful improvement in PsAID-12 scores at Week 4, which continued to Week 16 and was sustained to 1 year. Gossec et al. assessed 1-year bimekizumab efficacy from the patient perspective using the PsAID-12 questionnaire in bDMARD-naïve (BE OPTIMAL) and TNFi-IR (BE COMPLETE) patients with active PsA.

Deucravacitinib improved physical and social functioning, mental health, fatigue, and pain in a
Phase 2 trial in patients with active PsA. Here, investigators aimed to report the impact of deucravacitinib in a Phase 2 study in patients with active PsA from a patient perspective.

Significant improvements in overall disease activity, enthesitis and dactylitis, and skin psoriasis were observed by Week 8 and maintained or improved through Week 100 in both guselkumab treatment groups. Coates et al conducted a post-hoc analysis of the Phase 3 DISCOVER-2 trial to investigate the long-term (100-week) efficacy of guselkumab across GRAPPA-identified PsA domains.
