Vías de señalización intracelular: potencial para tratamientos

Current Rheumatology Reports 2009; 11:378-85

With recent progress in the development of drugs targeting signalling pathways for rheumatoid arthritis, this review article from the National Institutes of Health (NIH) provides an overview of the key intracellular pathways involved in the pathogenesis of rheumatoid arthritis. This paper also discusses some of the limitations of current drug targets including lack of clinical efficacy, potential adverse effects and cost, and highlights important issues associated with the design of target drugs such as the need for suitable animal disease models. The authors suggest that the structural similarity between kinases adds to the complexity of developing drug targets with acceptable safety and efficacy.
