Patient Clusters Identified by Machine Learning from a Pooled Analysis of the Clinical Development Programme of Secukinumab in Psoriatic Arthritis, Ankylosing Spondylitis and Psoriatic Arthritis with Axial Manifestations

Psoriatic arthritis clusters, obtained by machine learning (ML) analysis of pooled data from the FUTURE, MEASURE, and MAXIMISE trials, indicate phenotypical heterogeneity of patients with PsA and axial manifestations and overlapping features across the spondyloarthritis spectrum. Here, Baraliakos, et al. sort to identify distinct clinical clusters, based on patient demographics and baseline clinical indicators, from the secukinumab clinical development programme.

Bimekizumab Treatment in Biologic DMARD-Naïve Patients with Active Psoriatic Arthritis: 52-Week Efficacy and Safety Results from the Phase III, Randomised, Placebo-Controlled, Active Reference BE OPTIMAL Study

Data from this phase 3 RCT demonstrated that the efficacy of bimekizumab observed at 16 weeks remained consistent through to 52 weeks in the treatment of bDMARD-naïve patients with PsA. Patients who started the trial on placebo and switched to bimekizumab at week 16 showed similar improvements to those patients who were randomised to receive bimekizumab at the start of the trail. No new safety signals were identified.

Efficacy of Tofacitinib on Enthesitis in Patients with Active Psoriatic Arthritis: Analysis of Pooled Data from Two Phase 3 Studies

Post-hoc analysis of two tofacitinib phase three studies concludes that tofacitinib treatment resulted in improvements in enthesitis in patients with PsA, regardless of baseline location or severity.

Apremilast Long‑Term Safety Up to 5 Years from 15 Pooled Randomized, placebo‑Controlled Studies of Psoriasis, Psoriatic Arthritis, and Behçet’s Syndrome

This study confirms the safety of long-term apremilast use in patients with plaque PsO, active PsA, or oral ulcers associated with Behçet’s syndrome. In coming to this conclusion, investigators conducted a pooled analysis of apremilast data from 15 clinical studies with open-label extension phases, focusing on long-term safety.

Secukinumab Improves Physical Function and Quality of Life and Inhibits Structural Damage in Patients with PsA with Sustained Remission or Low Disease Activity: Results From the 2-year Phase 3 FUTURE 5 Study

Results from the 2-year phase 3 study FUTURE 5 show that the majority of patients with PsA who are treated with secukinumab were able to achieve sustained low disease activity or remission by week 104.

Guselkumab, a Selective Interleukin-23 p19 Subunit Inhibitor, Resolves Dactylitis in Patients With Active Psoriatic Arthritis: Pooled Results Through Week 52 From Two Phase 3 Studies

Post hoc analysis of guselkumab, Phase 3 DISCOVER-1 and -2 studies finds that 75% of guselkumab-randomised patients have complete resolution of dactylitis through one year.

Efficacy of Secukinumab on Dactylitis in Patients with Active Psoriatic Arthritis from the FUTURE 5 study

Low Rates of Radiographic Progression Associated with Clinical Efficacy Following up to 2 Years of Treatment with Guselkumab: Results From a Phase 3, Randomised, Double-blind, Placebo-controlled Study of Biologic-naïve Patients with Active Psoriatic Arthritis

This study sought to evaluate the relationship between radiographic progression and clinical outcomes in post hoc analyses of patients with PsA receiving up to 2 years of guselkumab therapy in DISCOVER-2.
