Lebwohl et al. investigated the efficacy and safety of risankizumab compared with placebo for treating palmoplantar psoriasis. At Wk16, significantly more patients receiving risankizumab achieved palmoplantar Investigator’s Global Assessment (ppIGA) 0/1 and PPASI75. The results were sustained through Wk52 with no new safety signals.

Renkhold et al. report that secukinumab significantly reduced psoriasis-associated pruritus intensity, improved skin lesions, and normalised histopathological changes, with stable neuroanatomy despite treatment discontinuation.

Tsai et al. conducted a systematic literature review and network meta-analysis evaluating deucravacitinib and other systemic treatments for moderate-to-severe plaque psoriasis in Asian populations. The authors reported that deucravacitinib achieved PASI75 and PASI90 response rates of 66% and 40%, respectively, higher than placebo and apremilast.

Tsai et al. observed no significant difference in the incidence of psoriatic arthritis among psoriasis patients treated with IL-23 inhibitors compared to IL-12/23 inhibitors, although there was a numerically lower PsA risk with IL-23 inhibitors. Results indicate both therapies are similarly effective for PsO management.

Poddubnyy et al. analysed five clinical trials to evaluate extra-musculoskeletal manifestations (EMMs) like uveitis, IBD, and psoriasis in patients treated with UPA. They observed low incidences across PsA, r-axSpA, and nr-axSpA. Numerically, uveitis rates were lower in
UPA-treated patients than in those receiving placebo, particularly in r-axSpA.

Ghani et al. compared the efficacy and safety profiles of tapinarof and roflumilast for treating mild-to-moderate plaque psoriasis. Both therapies showed robust efficacy and were well-tolerated, with low rates of adverse events. Tapinarof exhibited marginally higher efficacy in PASI scores compared to roflumilast.

Cai et al. demonstrated that xeligekimab significantly improved the Psoriasis Area and Severity Index (PASI) scores in patients with moderate-to-severe plaque psoriasis, with 90.7% achieving PASI 75 at week 12. Xeligekimab was well-tolerated with no unexpected safety concerns.

Following discontinuation of secukinumab 150mg or 300mg, a proportion of patients sustained low PASI with clear or almost clear skin despite being drug free for up to 2 years. Patients with a shorter disease duration were less likely to relapse, further supporting the hypothesis that earlier intervention with secukinumab may result in long-term control of moderate-to-severe psoriasis.

Kristensen et al. compared 14 PsA drugs across five treatment classes, evaluating their real-world effectiveness over three months. Ixekizumab showed rapid effectiveness on joint disease activity and skin outcomes, performing better than IL-12/23i and IL-23i, and comparable to TNFi and JAKi. More patients with active psoriasis achieved minimal disease activity with Ixekizumab than other therapies.

Results of this analysis by Blauvelt, et al. showed a low adjudicated suicidal ideation and behaviour (SIB) rate of 0.13/100 patient-years for bimekizumab, consistent with general psoriasis population ranges. Bimekizumab did not increase the risk of SIB compared to other anti-IL-17A/anti-IL-23 therapies.
